Each issue is designed to be thought provoking and inspiring, so that you can witness how the school's wisdom lights up the stars of quest for knowledge. From this issue onwards, we will focus our attention to experiments held in the Joint Research Center. The teachers from Subject Catalyst will come regularly to provide students with in-depth theoretical explanations and hands-on instructions, leading them to explore the mysteries of life sciences together.
本次课程,我们以“似‘魔法’的基因剪刀”为主题,带领同学们深入探索科学的神秘领域。生动的讲解和实验揭示了CRISPR技术如何像一把锋利的剪刀,精准地修剪和修补生物的DNA序列,展现这一技术在医学领域的巨大潜力。
In this course, with the theme of ‘Genetic Scissors’, we led students to explore the mysterious field of science. Vivid explanations and experiments revealed how CRISPR technology is like a pair of sharp scissors, precisely trimming and repairing biological DNA sequences, showing the great potential of this technology in the field of medicine.
Lily老师毕业于上海农业科学院和上海海洋大学,拥有生物学硕士学位,并在SCI期刊上发表了多篇论文。她参与了国家重点开发项目,研究领域广泛,包括微生物生态学、生物化学与分子生物学、植物学及生态环境监测。
Lily graduated from Shanghai Academy of Agricultural Sciences and Shanghai Ocean University with a master's degree in biology and has published several papers in SCI journals. She has participated in national key development projects in a wide range of research areas, including microbial ecology, biochemistry and molecular biology, etc.
课程在天津法拉古特学校·思楷联合实验室进行。该实验室有先进的生物实验室设备,让学生在高中就能接触大学水平的实验设施。活动当天,Lily老师的身影在实验室中忙碌着,她早早地开始准备,确保高速离心机、蓝光切胶仪、电泳仪、移液器等设备和物料一应俱全,证明了她对科学教育的热爱,也为学生们提供一个安全、高效的学习平台。
The course is conducted in a Joint Research Center created by our school and Subject Catalyst. The center has state-of-the-art biology lab equipment, giving students access to college-level lab facilities in high school. On the day of activity, Teacher Lily was busy in the center. She started her preparations early and made sure that the equipment and materials such as high-speed centrifuge, blue light cutter, electrophoresis, pipettes, etc., were all available, proving her love for science education and providing a safe and efficient learning platform for students.
理论讲解 Theoretical Explanation
Lily老师为学生们揭开了基因编辑的神秘面纱。她细致地讲解了CRISPR质粒的设计原理,以及如何通过大肠杆菌DH5a的转化,利用cas9和gRNA质粒精确剪切lacZ乳糖操纵子。这不仅让学生们对基因敲除技术有了深刻的理解,也让他们见证了“基因剪刀”CRISPR的神奇效果,课程的亮点在于将复杂的科学概念转化为学生能够直观感受和理解的知识。
Teacher Lily unravelled the mystery of gene editing for students. She explained in detail the design principle of CRISPR plasmid and how to precisely shear lacZ lactose manipulator using cas9 and gRNA plasmid through the transformation of E. coli DH5a. This not only gave students a deep understanding of gene knockout technology, but also allowed them to witness the magical effect of CRISPR, the ‘gene scissors’. The highlight of course is to transform complex scientific concepts into knowledge that students can intuitively feel and understand.
实验准备 Experimental preparation
Lily老师在传授基因编辑的理论知识后,特别强调了实验安全的重要性。她提醒学生们,尽管本次实验不涉及高风险生物材料,但为防止污染,学生们在操作时需佩戴手套或确保手部清洁。在她的细心指导下,学生们纷纷戴上手套,谨慎而兴奋地开始了他们的实验探索,这一环节不仅增强了学生们的安全意识,也为他们即将进行的实验操作提供保障。
After teaching the theoretical knowledge of gene editing, Lily particularly emphasised the importance of experimental safety. She reminded the students that although this experiment did not involve high-risk biological materials, to prevent contamination, they needed to wear gloves or make sure their hands were clean during the operation. Under her careful guidance, students put on gloves and cautiously and excitedly started their experimental exploration. This session not only enhanced students' safety awareness, but also provided a guarantee for their upcoming experimental operations.
在课程的实验操作环节,同学们在Lily老师的指导下,在超净工作台内手持移液器,以极度的细致与谨慎,用枪头精准地挑取了LB平板上生长旺盛、形态良好的单个菌落。这一步骤为后续的实验奠定了坚实的基础。学生们的动作小心翼翼,体现了对实验操作的尊重和对科学细节的追求。他们的专注和精确,展现实验科学中的严谨态度和对结果的期待。
In the experimental operation part of course, students, under the guidance of Lily, held the pipette in the ultra-clean bench, and with extreme care and prudence, picked the single colony with vigorous growth and good morphology on the LB plate precisely with the tip of gun. This step laid a solid foundation for the subsequent experiments. The careful movements of students reflected the respect for experimental operations and the pursuit of scientific details. Their concentration and precision demonstrated the rigorous attitude in experimental science and the expectation of results.
紧接着,同学们利用PCR技术对挑选出的菌落进行了扩增,以验证基因敲除是否达到预期效果。最终,通过对扩增后的菌株基因lacZ进行电泳分析,同学们清晰地观察到了电泳图谱上的条带变化。这一结果不仅验证了基因敲除的成功,更让同学们亲眼见证了“基因魔剪”的精准剪切能力,加深了他们对基因编辑技术实际应用的认识。
Immediately after that, students used PCR to amplify the selected colonies in order to verify whether the gene knockout achieved the expected effect. In the end, through the electrophoretic analysis of amplified strain gene lacZ, students clearly observed the changes of bands on the electrophoretic profile. This result not only verified the success of gene knockout, but also allowed the students to witness the precise shearing ability of the ‘Genetic Scissors’, which deepened their understanding of the practical application of gene editing technology.
在实验课堂的忙碌氛围中,生命科学社社长Emily同学的身影显得格外突出。她不仅全情投入到自己的实验中,还不忘关照其他社员,耐心地解答他们的疑惑。随着实验的圆满结束,同学们的热情并未随之消退,他们围绕着老师,热烈讨论着实验中的每一个细节,思维的火花在交流中不断迸发。同学们的科学探究精神和团队合作能力在此刻绽放。
In the busy atmosphere of joint research center, Emily, the president of Life Science Club, stood out. She not only devoted herself to her own experiment, but also took care of other members and answered their questions patiently. With the successful completion of experiment, the enthusiasm of students did not subside, they surrounded the teacher, enthusiastically discussing every detail of experiment, the spark of thinking in the exchange continues to burst. The students' spirit of scientific inquiry and teamwork ability blossomed at this moment.
在这次活动中,我亲眼目睹了借助 CRISPR 技术对大肠杆菌基因进行修剪操作,cas9如同精密的导航仪,引导RNA 精准定位并结合目标DNA,整个流程有条不紊,十分有趣。这让我深切感受到基因编辑的神奇,为我开启了一扇洞察生命微观世界的新视窗,使我对现代生物科技的魅力和未来发展的潜力有了更为直观且深刻的认知。
In this activity, I witnessed the trimming operation of E. coli genes with the help of CRISPR technology. Cas9 is like a precise navigator, guiding the RNA to locate and combine with the target DNA precisely, and the whole process is orderly and very interesting. The whole process was very interesting and orderly. It made me deeply feel the magic of gene editing, opened a new window to the microcosm of life, and gave me a more intuitive and deeper understanding of the charm of modern biotechnology and the potential of future development.
在最近的实验室活动中,同学们体验了基础实验仪器的操作,与科学的亲密接触,甚至连非社团成员也被多样的实验内容所吸引。我们体验了CRISPR技术的基本内容。作为一项获得诺贝尔奖的突破性技术,cas9系统相关的实验经历让同学们更加贴近科学前沿。我们期待未来能有更多机会,让大家在实验中不断学习与创新。
During the recent activity, students were able to experience the operation of basic experimental instruments and get in touch with science, and even non-members of club were fascinated by the variety of experiments. We experienced the basics of CRISPR technology. As a breakthrough technology that won the Nobel Prize, the experiments related to the cas9 system brought students closer to the frontiers of science. We look forward to more opportunities in the future to learn and innovate through experiments.
随着本次课程的圆满结束,学生们不仅学习了CRISPR技术的理论知识,还亲手进行了实验操作,体验了科学的魅力和挑战。我们期待下一次的实验活动能够带来更多的惊喜和发现,法拉古特将继续为学生们提供这样的平台,让他们在探索与合作中不断成长,怀揣着这份对科学的热爱和执着,勇敢地攀登科学的高峰,不断追求新知,勇于创新。
With the successful conclusion of this course, students not only learnt the theoretical knowledge of CRISPR technology, but also carried out experimental operations with their own hands, experiencing the charm and challenges of science. We look forward to more surprises and discoveries in the next experiments, Farragut will continue to provide students with such a platform, so that they can grow up in exploration and cooperation, with this love of science and persistence, bravely climb the peak of science, and constantly pursue new knowledge and innovation.
